This thesis improved husbandry and human immune-cell engraftment methods for gnotobiotic severe combined immunodeficient pigs. Bovine gamma globulin had little effect on human-cell viability and could replace porcine immunoglobulin, while expanded CD34-positive cell injections produced circulating human cells in 11 of 15 pigs and broader immune lineages, sometimes supporting near-complete thymus development. UID UCT-2112 temperature microchips supported animal monitoring.
