This study combined human genetics with mouse experiments to evaluate chondroitin sulfate as an extracellular-matrix target for type 2 diabetes. Twelve related human genes were linked to glycemic traits and diabetes risk, while chondroitin sulfate sharply lowered glucose in severely diabetic mice and improved glucose tolerance, insulin secretion, and insulin action in diet-induced obesity without hypoglycemia.
