This study examined how type I and type III interferons restrict emerging tick-borne orthoflaviviruses. Cell-culture and receptor-knockout mouse experiments showed that both interferon families control infection in cell-type-specific ways, while a CRISPR screen identified the endoplasmic-reticulum protein IFI6 as a key blocker acting after viral entry.
